1. Autoagglutination

Using a nutrient agar medium, test a colony with a drop of saline solution on a glass slide.

Apply the saline solution and then the colony, homogenize, and observe the results after shaking the slide for 30–60 seconds. If the result is positive (agglutination), serotyping cannot be performed. It is an autoagglutinating bacterial strain.

2. O Antiserum Group

The procedure is the same as that described in step 1, with the drop of saline solution being replaced by a drop of antiserum. O antigen mixtures are tested first. If they are positive, then testing for monovalent O antisera belonging to this group will be performed. When a result is positive, the isolated bacterium possesses the antigen corresponding to the antiserum.

3. Vi Antiserum Group

Use the anti-Vi antiserum instead of the saline solution and follow the same procedure as described in step 2.

4. H Antiserum Group

Use the antiserum mixtures, then the anti-H monovalent antisera instead of the saline solution and follow the same procedure as in step 2.

A phase inversion test can be performed in a second step. In this case, follow the instructions given in the product technical data sheet.

After results are obtained, refer to the standard value in force or consult the Kauffmann-White manual for interpretation according to the monovalent antisera that are agglutinated.
